In order to contribute to the elucidation of the mechanisms involved in DNA repair, specially when the protein XPG is involved, we are determining the mutation present in Brazilian patients diagnosed as XP-G. Also, we are constructing recombinant adenoviruses carrying the cDNA of xpg gene and the egfp reporter gene. This vector will be used in complementation assays with XPG-/- cells. These adenoviruses will be useful to verify the dynamic of this protein in the nucleus of proficient and deficient-DNA repair cells, upon irradiation with UV light or after the exposition to Methylene Blue, an agent that induces oxidative lesions.
