Laboratório de reparo do DNA


Nossas atividades são principalmente a pesquisas relacionadas a mecanismos de reparo de DNA e mutagênese em seres vivos. Muitas de nossas abordagens incluem estudos em células de mamíferos (e humanas) em cultura, onde tentamos aprender os mecanismos de tolerância e remoção de lesões em DNA, além de processos de sinalização de morte celular (apoptose) em células deficientes em reparo de DNA (em grande parte provenientes de pacientes com a doença genética xeroderma pigmentosum). Desenvolvemos vários vetores virais recombinantes (derivados principalmente de adenovirus) para transdução de genes de reparo de DNA, que são utilizados nesses estudos. Esses vetores têm sido usados também diretamente para transdução em linhagens de camundongos nocaute para genes de reparo de DNA, o que nos coloca na trilha para desenvolvimento de protocolos de terapia gênica tendo a pele como alvo. Recentement, estamos desenvolvendo sistemas de inibição da expressão gênica com interferência de RNA (“knock-down”), visando a investigação de mecanismos de interação entre os diferentes processos de reparo de DNA. Com o acúmulo de enorme quantidade de dados gerados em abordagens genômicas, incluindo nossa própria participação em projetos genoma, nós temos desenvolvido estudos de análise de evolução genômica, utilizando ferramentas de bioinformática. De fato, genes relacionados a reparo de DNA são altamente conservados e altamente apropriados para esses estudos, como indicado pelo nosso trabalho com reparo de DNA em plantas. Além disso, tendo em vista o conhecimento da sequencia completa da bactéria Caulobacter crescentus, nós estamos buscando investigar os genes involvidos em reparo de DNA destas proteobactérias do grupo alfa. Esse modelo bacteriano tem sido muito usado para estudos de diferenciação e ciclo celular em procariontes e a possibilidade de sincronizar essas bactérias pode ser útil para determinação de efeitos do ciclo celular nas funções de reparo.
